Abstract
Cellulase was produced under solid-state fermentation by Trichoderma viride ZY-1 using wheat bran and straw as a substrate.The ratio effects of the rice straw to wheat bran on cellulase production was studied.The results showed that at higher concentration of wheat,bran,viz.the qualitative ratio of the rice straw to the wheat bran was 0∶5 and 1∶4,after incubation for 48 h and pH remained at 4.5,the concentration of extracellular protein was the lowest,the concentration of reducing sugars was the highest,and activities of filter paper cellulase(FPase),carboxymethyl cellulase(CMCase),and β-glucosidase(β-Gase) were low.When rice straw was used as sole substrate and pH remained at 7,the concentration of reducing sugars was the lowest,but the concentrations of extracellular protein was high,the FPase,CMCase,and β-Gase activities were low.As the ratio of the rice straw to the wheat bran was 3∶2 and 1∶4,the activities of FPase and β-Gase reached the highest,i.e.5.01 U/g dried substrate and 4.6 U/g dried substrate,respectively.As the ratio of the rice straw to the wheat bran was 4∶1,and fermentation time was 72 h,CMCase activity reached the highest at 6.01 U/g dried substrate.
