Expression and significance of HIF-1α and HIF-2α in human renal clear cell cancer

Abstract

Objective To explore the expression and clinical significance of Hypoxia-inducible factors HIF-1α and HIF-2α,in clear cell renal cell carcinoma(CCRCC).Methods The expression of HIF-1α mRNA and HIF-2α mRNA was checked in 56 cases of primarily diverged renal carcinoma and 52 cases of normal renal tissues at transcription level by RT-PCR.The expression of HIF-1α and HIF-2α on the protein level was detected by Western-blot.Results The expression rate of HIF-1α mRNA in tissue specimen of CCRCC was 87.5%,increased more significantly than the control group(P0.05);expression rate of HIF-2α mRNA in tissue specimen of CCRCC was 94.6%,increased more significantly than the control group(P0.05).In the 56 CCRCC tissue specimens,52 cases expressed HIF-1α protein,the positive rate was 92.9%,increased more significantly than the control group(P0.05).In 56 casesed CCRCC tissue specimens,54 cases expressed HIF-2α,and the positive rate was 96.4%,increased more significantly than the control group(P0.05).Conclusions The expression of HIF-1α and HIF-2α on the mRNA and protein level possibly play an important role in the genesis and development of CCRCC.
