Abstract
Objective:To explore the effect of liposomal transfection of survivin antisense oligodeoxynucleotide(ASODN) on leukemic cell lines K562 cell proliferation and apoptosis.Methods:Anti-survivin phosphorothioate ASODN was synthesized and transfected into K562 cells by liposome,cell morphological transformation was observed with inverted microscope;The effect on cell proliferation was examined by MTT assay. Cell apoptosis was ditermined in situ cell apoptosis detection kit(POD). Survivin mRNA expression was determined by RT-PCR.Results:(1)Survivin ASODN inhibited K562 cells proliferation in a dose and time dependent manner. (2)Survivin ASDON can induce the cell apoptosis and the apoptotic index is in a dose dependent manner. (3)The expression of survivin mRNA significantly decreased after treatment with surviving ASODN.Conclusion:Survivin ASODN is able to inhibit the proliferation of leukemic cells lines K562 cell and induce the cell apoptosis, which might provide a new target for tumor gene therapy.
