Effects of "Yiqi Huayu Bushen Recipe" on ColI and ColII mRNA Expressions of Degenerative Intervertebral Disc Cells in Patients with Cervical Spondylotic Myelopathy

Objective:To investigate the effects of Yiqi Huayu Bushen Recipe containing serum on ColⅠ and ColⅡ mRNA expressions of the degenerative intervertebral disc cells in patients with cervical spondylotic myelopathy.Methods:The degenerative intervertebral disc tissues were obtained from the patients with cervical spondylotic myelopathy through surgery,and the primary cells were cultured by tissue mass method;the male SD rats of SPF level were chosen to prepare the drug serum.The degenerative intervertebral disc cells of the first generation were vaccinated in 96-well microtiter plates,divided into six groups,and cultured by low(5%),medium(10%) and high(20%) concentration serum containing Yiqi Huayu Bushen Recipe and corresponding concentration serum containing normal saline,respectively.The Col Ⅰand Col Ⅱ mRNA expressions were detected by real-time quantitative PCR method.Results:The different concentration serum containing Yiqi Huayu Bushen Recipe could obviously promote proliferation of the intervertebral disc cells.Compared with the corresponding concentration serum containing normal saline groups,the Col Ⅱ mRNA expression was increased,and the Col Ⅰ mRNA expression was decreased in different concentration serum containing Yiqi Huayu Bushen Recipe groups,especially the medium concentration group was more obvious(P0.01).Conclusion:Yiqi Huayu Bushen Recipe may regulate collagen expression in degenerative intervertebral disc cells to prevent and treat cervical spondylotic myelopathy.
