The influence of TNF-αand IFN-α on the function of Human Dendritic Cells

Abstract

Objective To evaluate the function of DC which stimulated by the different cell factors.Methods Peripheral blood monocyte-derived DCs progenitors were assigned to two groups:one was cultured with GM-CSF/IL-4/IFNα for 9 days(IFNα-DC group),the other with GM-CSF/IL-4/TNFαfor 9 days(TNFα-DC group).Cell morphology was monitored under light microscopy and cell surface molecules including HLA-DR,CD80,CD1α,CD83 were assayed by flow cytometry.The concentration of IL-12 in the supernatant were assayed by ELISA.T cell proliferation induced by DC was assayed by Methyl thiazolyl tetrazolium(MTT).Results Compared with two types of DC,IFNα-DC group were characterized as follows:①Increased secretion of IL-12;②High expression levels of CD1a,CD80,CD83;③Stronger activity of stimulating proliferation of isogenic T cells.Conclusion Compared with TNFα-DC group,to s certain extent,IFNα-DC group have higher function.
