Abstract

Objective To investigate the damage caused by high glucose, and to measure the role of hepatocyte growth factor(HGF) on fibroblasts. Methods Results Fibroblasts from the human kidney tumor were cul-tured. The cellular proliferation in media containing high glucose and normal glucose was assessed respectively using MTT. Flowcytometry was used to study the cell cycle. The role of exogenous HGF on cellular proliferation was also measured. Results After 36h , in comparison to the control group, high glucose concentration induced the change of cell cycle in fibroblasts. The cells were arrested in the G1 -phase of the cell cycle. At the same time high glucose in-hibited cellular proliferation. HGF modulated the proliferation of fibroblasts. Conclusion In vitro high glucose suppresses human fibroblasts proliferation and induces cellular hypertrophy. Exogenous HGF can prevent cell hyper-trophy.
