Abstract
Objective:To study the difference in gene expression between human TCC and normal epithelia tissue.Methods:Suppression subtractive hybridization technique was used to construct a cDNA subtractive library of human TCC,Dot blot was then used to screen the genes that differently expressed between TCC and it's normal epithelia cells.Some clones in the liˉbrary were selected randomly and sequenced.Then the results were analyzed with the BLAST software.Results:Human TCC subtractive library with high subtractive efficiency was set up successfully,which contained376positive clones.100clones were selected randomly to be amˉplified screened and89clones of them were differently expressed genes that were about200~900bp long.After sequencing.It was found that there were41kinds of known genes and5unknown genes in them.Conclusion:Studying the differently expressed genes in human TCC by SSH and Dot blot was a highly efficient way for screening and cloning new and specific genes.It also lays solid foundation for researching the function of genes and the mechanism of TCC.
