Abstract

Obiective To culture and identify human umbilical cord blood MSCs(mesenchymal stem cells)using three phase approach.Methods Through three phases(removal ofred blood cell,briefly culturing and phase of lasting culturing)to separate MSCs from 4 human umbilical cord blood samples.Culture medium with low glucose DMEM was supplemented with 20% fetal calf serum.Isolated MSCs were passaged to the third generation for identifying.Results Three groups of MSCs were separated from 4 groups of samples.By flow cytometry analysising,cells expressed MSCs surface markers of CD29,CD44 and CD105 positively and CD34 negatively.The result of the differentiation of osteoblast-like cells derived from human umbilical cord blood was positive also.Conclusion We can easily and effectively separate MSCs from human umbilical Cord Blood by three phases approach.
