Abstract
Objective To study the role of a cationic liposome, Geneshuttle20, on the uptake and distribution of STAT6 antisense oligonucleotide in murine spleen lymphocytes. Methods The uptake and intracellular distribution of STAT6 antisense oligonucleotide mediated by Geneshuttle20 were detected by flow cytometry and fluorescence microscopy. Results Compared with antisense oligonucleotide alone, combination of antisense oligonucleotide and cationic liposome in 2:4 w/w ratio showed a 6-fold rise (67.7%) in lymphocyte uptake, with highest intensity of intracellular fluorescence and nuclear staining. Conclusion Geneshuttle20 significantly enhance the uptake of STAT6 antisense oligonucleotide by murine spleen lymphocytes and facilitated its intranuclear effects.
