Effects of pyrrolidine dithiocarbamate on the proliferation,apoptosis and cell cycle in esophageal cancer EC1 cell line

Abstract

Aim:To investigate the effects of pyrrolidine dithiocarbamate(PDTC) on the proliferation,apoptosis and cell cycle in esophageal cancer EC1 cell line.Methods:Cell proliferation of EC1 treated by different dosage of PDTC(1,5,10,50,100 μmol/L) for 24,48 and 72 h was oberserved by MTT.The apoptosis and cell cycles of EC1 treated by different dosage of PDTC(0,5,10,50,100 μmol/L) for 48 h were measured by flow cytometry.The cells without treatment were the control.Results:PDTC inhibited the proliferation of EC1 cells in dose-dependent and time-dependent manner(Fconcentration=8.950,P=0.005;Ftime=30.718,P0.001;Finteraction=60.504,P0.001).After the treatment of PDTC for 48 h,cell apoptosis rate of EC1 cells increased with the dosage of PDTC increasing(F=7.020,P=0.001),the percentage of cells in G0/G1 phase decreased(F=771.064,P0.001),however,the percentages of cells in G2/M and S phase increased(F=963.002,309.332,P0.001).Conclusion:PDTC could inhibit the proliferation of EC1 cells,induce EC1 apoptosis and arrest cell cycle in S phase in vitro.
