Protective effect of platelet factor 4 on acute hematopoietic radiation injury in mice and its mechanism

Abstract

Objective: To investigate the protective effect and mechanism of platelet factor 4(PF4)on irradiated mice. Methods: One hundred male mice were randomized to several groups and exposed whole-bodily to 7.5 Gy{sup 60}Co {gamma}-rays. The mice were treated with injections of PF4(20 {mu}g/kg to 60 {mu}g/kg)twice at an interval of 6 h and 20 h, after the second injection they were irradiated. The 30-day survival rated were recorded. Results: The 30-day survival rate and the mean survival days, bone marrow cellularity. DNA content of bone marrow cells, number of CFU-GM on agar culture, peripheral blood cells, spleen/body weight ratio and number of spleen colonies were all significantly higher in the PF4 treated groups than in the corresponding irradiated controls (P<0.01). Conclusion: PF4 has beneficial protective effect on irradiated mice. Its mechanism could be related to protection of hematopoietic cells and acceleration of their recovery.
