Abstract
Objective:To elucidate the enhanced effect of IL3 on the proliferation and differentiation of cord blood hematopoietic cells in vitro.Methods:To preliminarily purify cord blood hematopoietic cells by continuiously double FicollHypaque method.The adult marrow stromal layer was establishied by static liquid culture,then irradiated with γray.The mononuclear cells(MNCs) were cultured in stromalcontact system with or without IL3 for six weeks.The nonadherent cells and CFCs were counted and measured at different time point.Results:The experimental group with IL3 was remarkable greater increase than control group in terms of the number of nonadherent cells(2.7~4.5 fold increase)and the production of CFCs(1.6~2.3 fold increase).Conclusion:IL3 enhanced the proliferation and differentiation of positive hematopoietic cells from cord blood in the stromasupported culture.
