Abstract
Objective To determine whether ursolic acid(UA) can induce apoptosis in human acute monocytic leukemia(THP-1) cell line and its mechanism. Methods THP-1 cells were treated with UA. The proliferative inhibition was examined by MTT method. HOECHST 33258 fluorescent staining and flow cytometry were used to detect apoptosis. Results UA inhibited the proliferation of THP-1 cells. THP-1 cells treated with UA were induced to apoptosis. Apparent morphological index of apoptosis was observed. G_ 1 phase declined, cells were mainly arrested in S phase. Conclusion Apoptosis of THP-1 cell is the key mechanism for UA’s killing action in the treatment of cancer.
