Abstract
Objective To observe the effects of different concentrations of sevoflurane postconditioning on inflammatory reactions in rats with cerebral ischemia-reperfusion injury following middle cerebral artery occlusion(MCAO).Methods Forty Wistar rats were randomly assigned to groups I V.Rats in the sham group(group S)were subjected to the vessels isolation without occlusion of the middle cerebral artery and exposed to 1.0MAC sevoflurane at 90min after surgery for 30min.Rats in ischemia-reperfusion group(group IR)were subjected to MCAO for 90min and then treated with O2for 30min at the beginning of reperfusion.Sevoflurane postconditing groups 1,2,and 3(groups PS1,PS2,and PS3)were subjected to MCAO for 90min,and then exposed to 0.5,1.0,or 1.5 MAC sevoflurane for 30min at the beginning of reperfusion,respectively.Blood samples were collected from femoral artery at 24hafter reperfusion,and serum TNF-α,IL-10,IL-1βwere measured. Results Compared with group S,the serum concentrations of TNF-αand IL-1βin group IR were increased,whereas the concentration of IL-10was decreased significantly(P0.05).Compared with group IR,serum concentration of TNF-α,IL-1βwere decreased and serum concentration of IL-10was increased in groups PS1,PS2,PS3(P0.05)in a dose-dependent manner.Conclusion Sevoflurane postconditioning not only exerts neuroprotective effect on inschemia-reperfused rats but also ameliorates inflammatory reactions.
