Abstract
Objective To investigate the mechanism of granuloctye colony stimulating factor(G-CSF) in the intervention of vascular dementia(VD) rats.Methods Fifty-four aged rats were randomly divided into sham group,VD model group and G-CSF intervention group.The VD model group and G-CSF group was set up with a permanent bilateral occlusion of both common carotid arteries for establishing vascular dementia models.The G-CSF intervention group rats were given G-CSF 50μg/kg every day after operation by hypodermic injection.Morris water maze test was conducted to observe the rats escape latency and evaluate the spatial learning and memory abilities.Western Blot was applied to detect the expression of Ach, CHAT in rat hippocampus.Results(1) The change of escape latency:compared with the sham group,the average duration of escape latency was prolonged in the groups of the VD model and G-CSF intervention(P0.01 ).Compred with the VD model group,the average duration was shorten afger G-CSF internvented(P0.01 ).(2 ) The expression of Ach,CHAT; compared with the sham group,the expression of Ach,CHAT in the hippocampus of the VD model group and the G-CSF intervention group was significantly increased at the time points of 7,14,28 days(P0.01 ).Compared with the VD model group,the expression levels were increased in the group of G-CSF interventional(P0.01).Conclusion G-CSF could promoted the expression of the Ach,CHAT in the hippocampus,and improve the learning and memory ability in vascular dementia rat.
