Determination of Chlorogenic Acid and Rutin Contents in Seabuckthorn Leaves by RP-HPLC

Abstract

A RP-HPLC method for simultaneous determination of chlorogenic acid and rutin in Seabuckthorn leaves was established.A Kromasil C18 column(150×4.6 mm,5 μm) was used with acetonitrile-1% acetic acid(20∶80) as the mobile phase,and UV-detection wavelength was set at 358 nm.The linear range was 0.0284 ~0.1704 mg·mL-1(r =0.9995) and 0.0566~0.3396 mg·mL-1(r=0.9992) for chlorogenic acid and rutin,respectively.The average recoveries(n=5) were 100.5% and 98.5%,respectively.This method is rapid,accurate with good reproducibility,which can be applied to the determination of chlorogenic acid and rutin in Seabuckthorn leaves.
