Abstract
Objective: To study the changes of dopaminergic neurons in mesencephalic ventral tegmental area(VTA) of Parkinson disease(PD) rats. Methods: 6-hydroxydopamine (6-OHDA) was injected into right substantia nigra compacta (SNc) of rats for making PD model. The praxiology changes induced by apomorphine were observed, neuronal alteration of left mesencephalic VTA were studied adopting electron microscope and Nissl's staining, and the changes of DA neurons were analyzed by tyrosine hydroxylase(TH) immunohistochemistry and image analysis. Results: The model rats showed abnormal rotational behavior, while neurons of mesencephalic VTA swelled and necrosed, and the amount of TH positive neuron decreased obviously. Conclusions: Mesencephalic VTA DA neurons take part in the pathologic changes of Parkinson disease(PD)rats. Abnormal rotations of model rat can be induced by APO injection, and the degree of rotation correlate with the amounts of TH positive neuron.
