Abstract

In order to build a genetic transformation system for the elite barley varieties in middle and lower reaches of the Yangtse River of China,the immature embryos of barley variety Supi 4 was used for explants to study the plantlet regeneration system.Compared with the treatment of 2.0 mg / L 2,4-D added to callus induction medium,2.0 mg / L Dicamba treatment significantly promoted the callus induction,and the callus induction frequency was increased from 78.33% to 100.00%.2.0 mg / L Zeatin(ZT) added to callus differentiation medium was the best for the callus differentiation,and the callus differentiation frequency was 75.56%.The differentiation promoting treatment for callus was better than the direct differentiation treatment,and the callus differentiation frequency was improved from 75.56% to 95.56%.The treatment of 0.5 mg / L malt extract(ME) in the root induction medium could significantly increase the number of root per plantlet.This plantlet regeneration system could be further used in genetic transformation and other genetic studies of barley.
