Inhibitory effects of Her-2/neu gene in SKOV3.ip1 cell by RNA interference

Abstract

Objective To study the inhibition of Her-2/neu gene in SKOV3.ip1 cell,a cell line of ovarian cancer, transfected with plasmid expressing short hairpin RNA(shRNA) targeting Her-2/neu gene. Methods After the SKOV3.ip1 cells were transfected with plasmid expressing shRNA,the expression of Her-2/neu gene was assessed in both mRNA and protein levels.Apoptosis of transfected cells were also detected to evaluate the biological effect of this method. Results RT-PCR and Western blot results indicated that the Her-2/neu gene exression was efficiently suppressed by plasmid expressing shRNA targeting Her-2/neu.Apoptosic rate of transfected cells were increased obviously. Conclusion The plasmid expressing shRNA targeting Her-2/neu can inhibit the expression of Her-2/neu gene effectively.
