Abstract

Objective: To study the inhibition of the expression of survivin by survivin small interterence RNA(siRNA) synthesized in vitro in osteosarcoma cell line U2-OS.Methods: Survivin siRNA was chemically synthesized in vitro and then transfected into cell line U2-OS.The expressions of survivin mRNA and protein both before and after transfection were detected by reverse transcription polymerase chain reaction(RT-PCR) and Western Blotting.The proliferation of cells was determined by MTT method.Results: Sequence specific siRNA targeting survivin down-regulated the expression of survivn mRNA and protein.The proliferation of cells was inhibited after transfection.Conclusion: Sequence specific siRNA targeting survivin can efficiently inhibit the survivin expression and cell proliferation in osteosarcoma cell line U2-OS.The successful application of survivin extends the list of available therapeutic modalities in the treatment of osteosarcoma.
