Abstract
The effect factors such as enzyme system,mycelium age,temperature and time of enzyme treatment on the formation and regeneration of protoplasts in strain Aspergillus niger Ni-5k were studied.The results showed that the suitable conditions were enzyme concentration 1.0%(containing 0.7% Cellulase,0.2% Snailase and 0.1% Lysozyme),mycelium age 18 h,zymolytic temperature 30°C,time 3 h.The protoplast yield was 10~6 cell/mL,and the regeneration percentage was 39% cultured with double layer medium in 32°C.
