Abstract

Objective To investigate the effect of carboxymethyl chitosan, hydroxyethyl chitosan, chondroitin sulfate, heparin and gelatin on the proliferation and morphology of human umbilical vein endothelial cells(HUVEC).Methods Cultured HUVEC in vitro, and then evaluated the effect of the above glycosaminoglycans and gelatin at different concentrations on the proliferation and morphology of HUVEC by MTT method. Results The above glycosaminoglycans and gelatin didn't inhibit the proliferation of HUVEC at the concentration range of 0~2 000 mg·L-1,and promoted the growth of HUVEC at certain concentrations within that range. The optimal concentration of carboxymethyl chitosan was 500 mg L-1,as well as hydroxyethyl chitosan, heparin and gelatin. The optimal concentration of chondroitin sulfate was 1 000 mg L-1.There was no obvious difference in cell morphology between each experimental group and the control group. Conclusion Carboxymethyl chitosan, hydroxyethyl chitosan, chondroitin sulfate, heparin and gelatin had good cytocompatibility with HUVEC,and promoted the proliferation of HUVEC at certain concentrations.
