Abstract

Objective To observe the influence of transfection KISS-1 gene in vitro on the ability of cell proliferation of gastric carcinoma BGC-823 cells and to explore the feasibility of using KISS-1 gene as therapeutic target for treatment of gastric carcinoma.Methods The eukaryotic expression vector with KISS-1 gene was transfected into BGC-823 cell line by using liposome -mediated transfection in vitro,the cells were selected by G418.The expression of KISS-1 gene was confirmed by Western blotting and reverse transcription polymerase chain reaction(RT-PCR).Methyl thiazolyl tetrazolium(MTT) was used to detect the ability of cell proliferation.Results The results of Western blotting and RT-PCR showed that the expression of KISS-1 protein and mRNA in cells transfected with KISS-1 were significantly higher than in cells transfected with pcDNA3.1 and control cells(P0.05);MTT showed that the proliferation ability of cells after transfection with KISS-1 was significantly reduced than cells transfected with pcDNA3.1 and control cells after 48 hours and 72 hours(P0.05).Conclusion KISS-1 gene may inhibit the proliferation ability of BGC-823 cells.
