Abstract
Objective To investigate the effects of Naomaitai on inflammation in focal cerebral ischemia/reperfusion rats.Methods Male Sprague-Dawley rats were randomized into six groups(n=10): sham-operated group,MCAO group,MCAO+ Naomaitai(2.24、 1.12、 0.56g/kg) group and MCAO+nimodipin(10mg/kg)group.Focal cerebral ischemia/reperfusion model in rats were made by transient occlusion of the middle cerebral artery for 90 minutes followed by 24 h reperfusion.At the 24th hour after reperfusion,the brains were obtained for TUNEL staining.NF-κB、 TNF-α、 IL-1β and ICAM-1 expression was detected by immunohistochemistry method.Results The activity of MPO and tile expression level of NF-κB、 TNF-α、 IL-1β and ICAM-1 in neuron increased successfully 24h after reperfusion.The protein expression of NF-κ B、 TNF-α、 IL-I β and ICAM-1 were significantly lowered in NaoMaiTai group than the MCAO group(P0.01).While the apoptotic cells were significantly decreased(P0.01).Conclusion These results showed that the anti-inflammatory protective effects of NaoMaiTai might be correlated with the decreased activity of MPO and inhibited the expression ofNF-κB、 TNF-α、 IL-1β、 ICAM.
