Abstract

OBJECTIVE: To investigate the effects of Xiaoliu formulaⅠon the apoptosis of MCF-7 human breast cancer cell, and to explore the molecular mechanism of the apoptosis of MCF-7 human breast cancer cell by analyzing the protein expression of Bax and Bcl-2 in MCF-7. METHODS: MCF-7 cells were treated with compounds of 1 μg·mL-1,10 μg·mL-1,100 μg·mL-1,1 000 μg·mL-1, and PI staining was conducted. Cell cycle was assayed by FACS, and Bax and Bcl-2 protein expression were analyzed by Western blot. RESULTS: The results of FACS showed that cells were retarded in G0/G1 phase after treating with Xiaoliu formulaⅠfor 24 h. Results of Western blot revealed that Xiaoliu formulaⅠimproved the protein expression of Bax and reduced protein expression of Bcl-2 with concentration-dependent manner. CONCLUSION: Xiaoliu formulaⅠcan induce the apoptosis of MCF-7 by regulating the protein expression of Bax/Bcl-2.
