Effects of Sodium Arsenite on Cell Viability,ROS Production and Apoptosis in Melanoma Cell

Abstract

Objective To observe the effects of sodium arsenite on cell viability,ROS production and apoptosis of human melanoma cell line(A375).Methods A375 cells were exposed to sodium arsenite at the concentration of 0,1,5,10 and 20 μmol/L for 24 hours.The cell viability was measured by Alamar Blue assay,and the levels of ROS and apoptosis were measured by flow cytometry.Results The cell viability of 10 and 20 μmol/L groups were significantly lower than that of control,while ROS levels were markedly higher than that of control;apoptosis level of 20 μmol/L groups was significantly higher than that of control.As the concentration of sodium arsenite increased,the cell viability decreased,while the levels of ROS and apoptosis increased.Conclusion Sodium arsenite can induce the increase of ROS production and apoptosis as well in human A375 cell.
