Isolation of one β-glucosidase gene in Aspergillus niger

Abstract

Genomic DNA and mRNA were isolated from Aspergillus niger, and the high efficient primers was designed by species similarity. With the methods of PCR and RT-PCR respectively, the fragments of the DNA and cDNA of beta-glucosidase gene were obtained. Seqence analysis indicated that DNA fragment was 2924 bp, and the cDNA sequence was 2583 bp, which encode 860 amino acids. The identity of the derived protein with Aspergillus niger β-glucosidase has the high homogeneous characteristics of 97%~99%. The foundmental researches for transformation and application of this gene have been achieved in this paper.
