Abstract

Objective To explore the feature of changes of event-related potential P300 and the effects of donepezil hydrochloride in vascular dementia mice with cognitive function change.Methods Forty-eight mice were randomized to sham operation group,vascular dementia model group and treatment with donepezil hydrochloride group.Each group included 16 mice.The mice were subjected to ischemia and reperfusion repeatedly by the ligation of bilateral common carotid arteries to establish the VaD models.They were treated with donepezil hydrochloride for 4 weeks.The behavioral changes were investigated by step-down test and water maze test.N2 and P3 were determined in mice with vascular dementia treated with donepezil hydrochloride and compared with those in the sham operation group and VaD model group.Results Compared with the sham operation group and mice with vascular dementia treated with donepezil hydrochloride,the learning records of model group mice were that the response time was longer,swimming time was longer and the error frequency was increased.Memory records of model group mice by step-down test were that latent time was shorter and in water maze test,the swimming time was longer and the error frequency was increased(P0.05).N2 and P3 potential latencies of model group mice were significantly longer than those of VaD mice treated with donepezil hydrochloride and sham operation group mice(P0.01);P3 amplitude was lower than that of the VaD mice treated with donepezil hydrochloride and sham operation group mice(P0.05).Conclusion Donepezil hydrochloride can intervene and treat damage of the cognitive functions in mice with vascular dementia
