Abstract
Objective:To explore the changes of cAMP-response element binding protein in the hippocampus of rats with Alzheimer′s Disease and observe the effect of electro-acupuncture therapy on the hippocampus.Methods:Male SD rats in number 40 were randomly divided into 4 groups in average,the normal group,the sham group,the model group,and the EA group.By utilizing condensed Aβ25~35 in one-time injection of bilateral hippocampus to induce AD model rats,we selected bilat-eralm Shenshu(BL23),bilat-eralm Neiguan(PC6) and Dazhui(GV14)acupoints in EA group.G6805-Ⅱ type of electroacupuncture treatment device was connected to needles,and then we selected the frequency of 2 Hz continus wave(CW),the intensity of 1 mA.The therapy last 20 mins,qd,6 times a week,a total of 2 weeks treatment.The expression of CREB protein in hippocampus was assayed by immunohistochemistry.Results:Compared with the sham group,the expression of CREB in hippocampus CA1、CA3 zone of the model group was lower,and the difference was significant(P0.01);compared with the model group,the expression of CREB in CA1 zone of the EA group was significantly increased(P0.05).The expression of CREB in CA3 zone of the EA group was increased,but there was no statistical significance(P0.05).Conclusion:Its mechanism may be achieved through the following mean: increasing the expression of CREB in the hippocampus,enhancing LTP,in order to improve the learning-memory capacity of rats with AD.
