Abstract
Objective:To investigate the apoptosis of bladder cancer cell BIU-87 induced by apigenin in vitro and its mechanism. Methods:The cell proliferation was detected by MTT assay;the cell cycle was measured by flow cytometry;the cell apoptosis was observed by flow cytometry with Annexin V-FITC/PI double staining;the expression of survivin mRNA was detected by RT-PCR,and the expressions of survivin protein and caspase-3 protein were detected by immunocytochemistry SP assay.Results:MTT showed that apigenin effectively inhibited the proliferation of BIU-87 cells.②After 72 hours of 22.03μmol/L apigegin treatment,it could arrest the BIU-87 cell in G2/M phase which was measured by flow cytometry,and it could make the cell apoptosis which was showed by flow cytometry with Annexin V-FITC and PI.③Compared with the control group,the expression of survivin mRNA was significantly decreased (P0.05),andsowas the expressionofsurvivinprotein(P0.05),while thatofcaspase-3 protein was marklyupregulated(P0.01)in the apigenin group.Conclusion:Apigenin inhibits the proliferation and induces the apoptosis of BIU-87 cells.The molecular mechanism of apigenin inducing the apoptosis of BIU-87 cells may be associated with its action of down-regulating the expression of survivin,and up-regulating the expression of caspase-3.
