Abstract
A SMART cDNA library was constructed by using the leaves of 98-160 stressed for 72 hours.Quality detection results showed that the titer of the unamplified cDNA library was 3.44×10~6 pfu/mL(white plague ratio85?%),the insert varied from 0.5 to 1.5?kb with the average of 800 base pairs,some were even larger than 2?kb.Therefore the cDNA library constructed according to the above method was a high-quality cDNA library which could be used for further full-length gene screening and cloning efficiently.
