Effects of Olfactory Ensheathing Cells on the Differentiation of Neural Stem Cells to Cholinergic Neuron

Abstract

Aim:To investigate the effects of different concentration of olfactory ensheathing cells(OECs) on the differentiation of neural stem cells(NSCs) to cholinergic neurons,and provide a theoretical basis for cell transplantation for the treatment of AD.Methods:OECs and NSCs were separately cultured in vitro.5×10~4/ mL NSCs and 1×10~4/mL,1×10~6/mL,1×10~8/mL OECs were co-cultured,respectively in 7 days.NSCs in the control group were left intact.ChAT positive cells were detected with immunocytochemical method. Results:After co-cultured for 7 days,it was found OECs could promote the differentiation of NSCs into cholinergic neurons.NSCs and 1×10~6/mL OECs group was more than the other groups.The increase in the differentiation of NSCs to cholinergic neurons showed significant statistic difference compared with the control group(P0.01).Conclusion:OECs can induce NSCs differentiate into cholinergic neurons.
