Correlation Among TNF-α、TNF-γ、IL-2 and Apoptosis of Pancreatic Islet Cells in Diabetes Mellitus Rat Model

Abstract

Objective To investigate the correlation between cytokines and apoptosis of pancreatic islet cells in rat model with Diabetes Mellitus(DM).Methods There were normal control and DM groups with 8 rats in each group.CK were measured and 6 rats in each group were tested for apoptosis study.Differences of CK level in plasma and pancreas,and mRNA expression in pancreas were analysed statistically.Pancreatic islet cells were separated by bile duct-digestion method.Apoptosis of islet cells were detected by the semi-quantitative analysis.The relationship between cytokines and apoptosis of pancreatic islet cells was analyzed with Person correlation method.Results The levels of TNF-α and mRNA expression in pancreas were increased significantly in DM group(P0.05).Ratio of apoptosis was elevated significantly in rats with DM[(17.52±9.95)% vs(8.45±2.01)%,P0.05].There were correlations between ratio of apoptosis and TNF-α level,as well as mRNA expression in pancreas.Conclusions TNF-α may induce apoptosis of pancreatic islet cells in rat model with DM directly.
