Dose effect of PDLLA/NGF compound conduit on restoration of rat sciatic nerve defect

Abstract

Objective To investigate the dose effect of poly d, l-lactic acid/nerve growth factor (PDLLA/NGF)compound conduit on restoring rat sciatic nerve defect. Methods The sciatic nerve defect of rats was bridged by PDLLA conduit, PDLLA/NGF compound conduit with 400μ NGF, and PDLLA/NGF compound conduit with 800 μ NGF. The results were evaluated in 3 months following operation by using electrophysiological and histological assessment. Results Nerve regeneration in PDLLA/NGF compound conduit with 400μ NGF was significantly superior to that in the other 2 groups. Conclusion NGF in PDLLA/NGF compound conduit has a dose-effect relationship on the repair of sciatic nerve defect. Suitable dosage of NGF can promote nerve regeneration, while too high dosage can inhibit nerve regeneration.
