The Expression of the Green Fluorescence Protein in Human Degenerative Nucleus Pulposus Cells Mediated by Adenovirus

Abstract

Objective Human degenerative nucleus pulposus cells were cultured in monolayer, the biological action of the cells was observed. And the green fluorescence protein gene was tranformed into the cells, and the expression of the exogenous marker gene was detected. Methods Human degenerative nucleus pulposus cells were cultured to set up a cell culturing model, the cells viability was assessed. The GFP gene mediated by adenovirus was tranformed into the cells to observed whether they could give out green light. Results The cells cultured in vitro increased slowly. With the cells culturing extended, the viability of the cells declined. The green light could be observed 24 hours after the GFP gene tranformed into the cells and the duration was up to 4 weeks. Conclusion The growing ability of the nucleus pulposus cells is feeble. The successful expression of GFP gene demonstrates that the exogenous gene can be expressed in human degenerative nucleus pulposus cells.
