Abstract
Objective:To investigate the effect of casticin on the proliferation of gastric cancer cell line SGC-7901,and explore the regulation of Bcl-2 via casticin.Methods:Pretreated the gastric cancer cell line SGC-7901 with casticin in different dose(1 g/mL,2 g/mL,4 g/mL,8 g/mL)and time(6h,12h,24h,48h),and proliferation of cells were examined by MTT;then Bcl-2 protein and transcriptional level were measured by Western blotting and RT-PCR after treated with casticin at concentration of IC50.Results:The cell proliferation was significantly inhibited when treated with casticin at 5.6 g/mL after 12 hours.The result of Western blotting and RT-PCR showed that Bcl-2 was down-regulated.Conclusions:Casticin repressed the proliferation of gastric cancer cell line SGC-7901,which was closely related to the down-regulation of Bcl-2.
