Abstract

Objective To establish insulin resistance (IR) rat model induced by a special diet.Methods 30 healthy male Wistar rats were randomly divided into the normal control group and the experimental group.The rats in the experimental group were fed with high-fat diet.IR were determined through the level of blood glucose,insulin and glucose infusion rate (GIR).Lipids levels and pathological changes of the liver and the terminal ileum were compared between the two groups.Results The weight gain in the experimental rats was significantly more than the normal control group(476±39)g vs.(425±21)g,P0.05.Compared with the normal control group,the levels of fasting plasma glucose(FPG) and fasting insulin(FIN) in experimental group were extremely significantly increased(7.2±0.6) vs.(4.8±0.1)mmol/L;(66.3±24.2) vs.(36.3±19.8)mmol/L,P0.01;the insulin sensitivity were significantly lower(8.1±1.1) vs.(18.2±2.1)mg·kg-1·min-1,P0.01;And serum total cholesterol,triglycerides,low density lipoprotein concentrations were also significantly higher than the normal group (P0.05).At the same time,hepatic steatosis and cytoplasm of osteoporosis in the experimental group appeared as well as necrotizing enterocolitis of terminal ileum.Conclusions IR rat model can be induced by high-fat diet and the appearance of visceral injury was related with IRS.
