Abstract
Objective:To observe the effects of neural stem cells(NSCs) transplantation on the proteolipid protein after the spinal cord injury(SCI) of rats,and to investigate the mechanism of myelin regeneration after SCI by NSCs transplantation.Method: Neural stem cells were cultured from the hippocampus of rat's embryo and identified by immunocytochemistry of Nestin.Then NSCs were labeled with Brdu (5-bromodeoxyuridine).Seven days after the operation of SCI the author transplanted the NSCs into the injured site.Sixty adult Wistar rats were randomly divided into three groups: spinal cord injury cured with transplantation of NSCs to injuryed site (group A),spinal cord injury received DMEM solution (group B),control group(group C).Then the NSCs labeled with Brdu and the expression of PLP were tested by Immunohistochemistry and RT-PCR.By means of above,the author investigated the mechanism of repairing the myelin after the NSCs transplantation.Result: NSCs can be detected in the spinal cord after transplantation.Compared with group B,transplantation of NSCs enhanced the expression of PLP at different level in the spinal cord of group A,improved the myelin regeneration.Conclusion: The transplantation of NSCs can repair the injuried site and promote the regeneration of myelin by enhancing the expression of PLP.It is one of the mechanism of repairing the myelin after SCI by NSCs transplantation.
