Abstract
Objective To study the reversal effect of resveratrol on multidrug-resistance of human breast cancer cell line MCF-7/ADM,and the related mechanism. Methods MTT assay was used to detect the inhibitory effect of resveratrol of different concentrations on the proliferation of human breast cancer cell line MCF-7/ADM,and the Jin's formula to assess the combined effect of Res and ADM on cancer line MCF-7/ADM.In addition,the concentrations of ADM in MCF-7/ADM cells were detected by Fluorescence Spectrophotometry while Resveratrol of different concentrations was given. Results Resveratrol combined with ADM possessed synergistic effect,and the reversal index was 1.950 in ADM+ 4 μmol/L Res,2.178 in ADM+ 8 μmol/L Res,and 2.355 in ADM+ 12 μmol/L Res.Resveratrol was observed to greatly elevate the concentration of ADM in MCF-7/ADM cells at a dose-related manner(compared to the control group,P0.05).Conclusions Resveratrol has a sensitizing effect on multidrug-resistance of human breast cancer cell line MCF-7/ADM in that it could partially reverse MDR in MCF-7/ADM cells.The reversal is coorelated with elevated concentration of ADM in MCF-7/ADM cells.
