An experimental study on thrombosis imaging with ~(99)Tc~m-H-Annexin V

Abstract

Objective This study evaluated the affinity of recombinant human Annexdn V(H-Annexin V)to thrombus,the labeling rate,stability of different Na~(99)Tc~mO_4 labeling methods,and the usefulness in imaging experimental thrombosis.Methods Annexin V,H-Annexin V and hydrazino-nicotinamide(HYNIC)- Annexin V were labeled with fresh Na~(99)Tc~mO_4 under 3 react conditions,then the labeling rate and stability of the resulted compounds were analyzed,with regional and whole body imaging performed at various times after injection in rabbit arterial thrombi models.The uptake ratio between injured to normal arteries was also calculated.Results In case of enough SnCl_2,~(99)Tc~m-H-Annexin V was simple and easy to control meth- ods.The labeling rate was95% and the radioehemical purity was80% after 17 h incubation at room temperature.In thrombus imaging,the ~(99)Tc~m-H-Annexin V showed higher tissue uptakes in injured artery. Conclusion ~(99)Tc~m-H-Annexin V has strong affinity to fresh thrombus,thereby it could be a good method in detecting arterial thrombus.
