Abstract
Objective To investigate the effect of spironolactone and valsartan on expression of apoptosis -associated protein P53,Bax and Bcl-2 in left ventricular myocytes of renovascular hypertension rats.Methods Forty-six male SD rats were randomized to hypertension group(group N, n = 12),spironolactone group(group S,n = 12) ,valsartan group(group Ⅴ,n = 11) ,spironolactone and valsartan group(group S+V,n = 11) ,control group(group C,n = 10).After 10 weeks of treatment,echocardiography was performed and carotid artery blood pressure was measured. Immunohistochemistry was adopted to examine the protein expression of P53,Bax and Bcl-2.Results Affer treatment for 10 weeks,compared with group Cthe blood pressure was higher in group N (P 0.01).Compared with group N,the blood pressure of groups Ⅴ and S+V was significantly decreased (P 0.05).Compared with group CBax,Bcl-2 and Bax/Bcl-2 were significantly increased in group N,Bax and Bax/Bcl-2 in group S were significantly increased (P 0.05,P 0.01).Compared with group N,Bax,Bax/Bcl-2 in group S and Bax,Bcl-2 ,Bax/Bcl-2 in groups V and S+V were significantly decreased.Compared with group S,Bax and Bax/Bcl-2 in groups V and S+V were significantly decreased (P 0.05,P 0.01).Conclusions Treatment with valsartan and spironolactone can decrease the expression of P53 and Bax in myocardium,inhibit the occurrence and partly reverse the LVH of renovascular hypertension rats.
