Abstract
Objective To observe the effect of atorvastatin on the expression of integrin α3β1 in the kidney of diabetic rats. Methods Twenty-one SD rats were randomly divided into three groups: normal rats,diabetic rats,diabetic rats treated with atorvastatin(20 mg·kg-1·d-1) for 8 weeks.Blood glucose,blood lipid,serum creatinine,urinary albumin excretion and Ccr were measured after 8 week's treatment;the expression of integrin α3β1 was determined by immunochemistry;the level of lipid peroxidatin(MDA) and total antioxidative capacity(TAOC) were examined by colorimetric method;the renal tissue pathology was observed under light microscope;In addition,the ultrastructure of glomeruli was observed by electron microscope. Results No significant differences of blood glucose and blood lipid were found between the diabetic rats and the atorvastatin treated rats;Compared with those of the diabetic rats,the level of Ccr,urinary albuminin excretion and the renal MDA of the treatment rats were decreased significantly,but the activities of renal TAOC and the expression of integrin α3β1 were increased markedly;renal pathologic changes were also improved Conclusion Atorvastatin attenuates proteinuria,improves renal pathology and protects renal function in the early stage of diabetes though decreasing the level of reactive oxygen species and up-regulating the expression of the integrin α3β1.
