Abstract
Objective To study the effect of umbilical cord blood mesenchymal stem cells on T lymphocytes.Methods UC-MSC was separated from umbilical cord.The purity of MSC was analyzed by flow cytometry;the proliferation of allogeneic T lymphocytes was measured by Brdu method;the CD69 expression was detected by flow cytometry.Results The immunophenotypic analysis showed that UCB-MSCs expressed no HLA-Ⅱ;UCB-MSCs could suppress umbilical cord blood lymphocytes proliferation stimulated by mitogen in dose-dependent manner;CD69+suppopulation was decreased(P0.01).Conclusion UCB-MSCs can supress T cells proliferation and activation,which indicates UCB-MSC might have negative immunomodulation and be useful in the prevention of GVHD in allogeneic stem cells.
