Abstract
Objective To construct the recombinant baculovirus containing HBV core gene in baculovirus expression vector system. Method HBV core gene was amplified by PCR from plasmid pHBV1 which contained HBV whole DNA sequence. The PCR product was cloned into pGEM- T vector, the gene of interest was inserted into the baculovirus expression vector pAcGHLT-A. After cotransfecting the cultured Sf9 cells with the recombinant plasmid pAcGHLT-HBcAg and the DNA of AcNPV respectively, the recombinant virus was obtained. Results The recombinant baculovirus containing the HBV core gene had been constructed. After infected Sf9, the correct fragment was amplified by PCR. Conclusion The recombinant baculovirus containing HBV core gene is constructed successfully by using baculovirus expression system, which lays the foundation for further research.
