Effect of ATRA on the expression of Caspase-3 in human lung adenocarcinoma cell line A549 and cell proliferation

Abstract

Aim: To investigate the effect of ATRA on the proliferation of lung cancer cell and expression of Caspase-3 in lung cancer cells.Methods:The effects of ATRA on the proliferation of lung cancer cells were measured by MTT method. Hoechest33258 fluorescent dyeing was used to observe ATRA-induced morphological changes. The expression of activated Caspase-3 protein was detected by Western blot.Results: Cell proliferation was significantly inhibited by ATRA(P0.05), the morphological changes of apoptosis such as nuclear fragmentation or condensed particles were identified after incubation with ATRA, and the expression of activated Caspase-3 protein was elevated.Conclusion: ATRA can inhibit the proliferation of lung cancer cell A549 and induce cell apoptosis by upregulating the expression of activated Caspase-3 protein.
