Abstract

Objective:To study the situation of T cell subpopulations in human umbilical cord and adult peripheral blood. Methods:Full blood differential counts were determined by blood routine examination. Stain the blood cells with fluorescent labeled anti-CD4/ anti-CD25/ anti-CD3 and anti-CD4/ anti-CD8/ anti-CD3 monoclonal antibody. Samples were run and analyzed for three-color immunofluorescence by flow cytometry. Results:The contents of erythrocytes、leucocytes、haematoglobin and the percentage of lympholeukocyte in umbilical cord blood were markedly differed from those in adult peripheral blood. The absolute count and the percentage of CD3+T cells、CD3+ CD4+ T cells、CD3+ CD8+ T cells、CD4+ CD8+ T cells、CD4+ CD25high regulatory T cells、CD4+ CD25int T cells and CD4+ CD25- T cells were markedly differed from those in adult peripheral blood. Conclusion -. The full blood differential counts and the T cell subpopulations in cord were markedly differed from those in adult peripheral blood. This result is probably corresponding with the immune state of the neonates.
