Abstract

Objective:To investigate the effects of insulin on human umbilical vein endothelial cells(HUVECs) induced by high glucose and analyze the expression changes of bcl-x and bax. Methods:HUVECs were isolated and incubated at different concentration of glucose and insulin for 72 hours, and total RNA was extracted.Using semi-quantitative RT-PCR techniques,we measured the expression of bcl-xL and bax mRNA in HUVECs.Results:High glucose could reduce the expression of bcl-xL and increase the expression of bax. After adding physiological or high concentration of insulin, the expressions of bcl-xL increased and the expressions of bax reduced. Conclusion:Insulin can increase HUVEC expression of bcl-xL and reduce the expression of bax.
