Abstract
Objective: To evaluate the effect of adenoviral vector containing bone morphogenetic protein-2 gene(Ad-BMP-2) on the osteogenic differentiation of human mesenchymal stem cell(MSC).Methods: MSCs were sampled from the iliac bone of young men and cultured.After bone morphogenetic protein-2(BMP-2) gene transfected into MSC,the expression of BMP-2 gene in MSC was determined by reverse transcriptase polymerase chain reaction(RT-PCR) and immunohistochemical staining,and the osteogenic activity of MSC was determined by in situ hybridization and immunohistochemical staining.Results: The expression of BMP-2 gene in MSC was stable after transfection.The expressions of core-binding factor a1(CBFa1),osteocalcin,osteopontin,alkaline phosphatase,and collagen type I were positive.The collagen excretion and calcium salt deposition were found under transmission electron microscope.Conclusion: Ad-BMP-2 transfection may promote osteogenic potential of MSC in vitro.
