Abstract
OBJECTIVE: To investigate the expression of CD147 siRNA on the expression of CD147 and the proliferation of lung adenocarcinoma cell line A2.and study the biolgical effects of targeting CD147 RNA interference on lung adenocarcinoma cell line.METHODS: CD147 siRNA was transfected into lung adenocarcinoma cell line A2 by LipofectamineTM2000,CD147 expression was monitored by RT-PCR,and the proliferation of the cells by MTT assay.RESULTS: After the interference of CD147 siRNA,the mRNA expression of CD147 in the A2 cells was significantly down-regulated.The proliferation levels of transfected A2 cells at 24,48 and 72 h after the interference decreased significantly by 48.3%,51.8% and 53.2%(P0.05).CONCLUSIONS: CD147 siRNA can effectively inhibit the expression of CD147 in A2 cells,and inhibit the proliferation of the cells.It is demonstrated that targeting CD147 RNA interference can potentially be a new approach for lung adenocarcinama gene therapy.
