Effect of Donepezil on Expression of Brain-derived Neurotrophic Factor in Hippocampus in Rats Induced by Aβ25-35

Abstract

Objective To explore the effect of donepezil(DN)on the expression of brain-derived neurotrophic factors(BDNF)in hippocampus of rats induced by Aβ25-35.Methods Forty-eight Wistar rats were divided randomly into groups control,DN and model,8 in each.Ad animal models were established by stereotactically injecting Aβ25-35 into the lateral ventricle.DN group were injected with DN for 4 weeks.The expression of BDNF in hippocampus was also determined by Western blotting.Results The expression of BDNF decreased in model group when compared with control group,(P0.01),and increased in treatment group as compared with model group(P0.05).Conclusion BDBF expression decreases significantly in rats injected with Aβ25-35,and increases in intervention group as compared with model group.Donepezil hydrochloride may increase BDNF expression in hippocampus of AD model rats.
